Stemness of Human Pluripotent Cells: Hypoxia-Like Response Induced by Low Nitric Oxide

The optimization of conditions to promote the stemness pluripotent cells in vitro is instrumental for their use advanced therapies. We show here that exposure human iPSCs and ESCs low concentrations chemical NO donor DETA/NO leads stabilization hypoxia-inducible factors (HIF-1α HIF-2α) under normoxia, with this effect being dependent on diminished Pro 402 hydroxylation decreased degradation by proteasome. Moreover, master genes pluripotency, NANOG OCT-4, were upregulated. also induces a shift metabolic profile PSCs, an increased expression hypoxia response glycolysis. Furthermore, reduction mitochondrial membrane potential lower oxygen consumption fusion regulators, such as DRP1, was observed. results reported indicate mimics PSCs enhances properties when cultured normoxic conditions.
